摘要:
      目的 明确背根神经节(DRG)中感觉神经元分泌的小外囊泡(sEVs)能否在体外影响后纵韧带细胞骨化。 方法 从手术标本中获得后纵韧带骨化症(OPLL)患者的韧带组织,提取韧带细胞并鉴定。提取纯化DRG中感觉神经元细胞分泌的sEVs,鉴定后作为刺激物与韧带细胞共培养。CCK8检测韧带细胞增殖情况,判断sEVs对韧带细胞活性的影响;在成骨诱导过程中加入sEVs,观察其对韧带细胞成骨分化的影响,并检测成骨标志物Runt相关转录因子2(RUNX2)和骨钙素(OCN)的表达。 结果 DRG中感觉神经元能够分泌sEVs,在sEVs刺激后,后纵韧带细胞的增殖能力显著上升,在成骨分化的过程中sEVs能够促进后纵韧带细胞成骨分化,并且促进成骨标志物RUNX2和OCN的表达。 结论 DRG中感觉神经元分泌的sEVs能够促进后纵韧带细胞骨化。

Abstract:
      Objective To investigate whether small extracellular vesicles(sEVs) secreted by sensory neurons in the dorsal root ganglion(DRG) can affect ossification of posterior longitudinal ligament(OPLL) cells in vitro. Methods Ligament cells were extracted and identified from ligament tissues obtained from surgical specimens of patients with OPLL. The sEVs secreted by sensory neurons in the DRG were extracted,purified and identified,and then co-cultured with ligament cells as a stimulus. The ligament cell proliferation was detected by CCK8,the effect of sEVs on the vitality of the ligament cells determined. The sEVs were added to the ligament cells during the osteogenic differentiation process,to observe their effect on the osteogenic differentiation of ligament cells,and the expression of the osteogenic markers of runt-related transcription factor 2(RUNX2) and osteocalcin(OCN) were detected. Results In DRG,sensory neurons could secrete sEVs. After stimulation with sEVs,the proliferation ability of posterior longitudinal ligament cells significantly increased. During the process of osteogenic differentiation,sEVs could promote the osteogenic differentiation of posterior longitudinal ligament cells and promote the expression of osteogenic markers RUNX2 and OCN. Conclusion The sEVs secreted by sensory neurons in DRG can promote the ossification of posterior longitudinal ligament cells.
